Spencer was born on March 9, 1975, in Tahlequah, Oklahoma. He grew up moving between several towns, including Kooskia, Lapwai, and Lewiston in Montana and Idaho. His heritage is diverse. He is of Lakota Sioux and Nez Perce descent, and also has connections to Cherokee, Muscogee (Creek), French, and Dutch ancestry.

Although he comes from a middle-class Indigenous family with both parents working in education and two younger sisters, Spencer is a well-known Indigenous activist and an enrolled member of the Fort Peck Assiniboine and Sioux Tribes. He holds American nationality.

Chaske's father works as a high school counsellor, and his mother is a teacher. Spencer attended Clearwater Valley High School and graduated in 1994. During high school, he performed under the name James Spencer at the Lewiston Civic Theatre.

He later enrolled at Lewis–Clark State College to study photography but left after one year to move to New York City and pursue an acting career.

His TV appearances include Barkskins on NatGeo, Blindspot on NBC, Jessica Jones on Netflix, the award-winning Cinemax series Banshee, as well as Longmire, Sneaky Pete, and The Society, where he played Mr Pfeiffer.

He then lost his girlfriend, his job, and even his lavish $3,000 per month apartment, and was required to move to a cramped, dirty location in eastern Brooklyn. He attended rehabs and, after becoming sober, Chaske was even on the verge of giving up acting.

After getting sober, Spencer considered stepping away from acting before receiving an audition opportunity for Twilight: New Moon, which became a turning point in his career. He received a call from his agent asking him to audition for Twilight's sequel titled "New Moon."
